Comparative analysis of the karyotype and meiosis in two species of Adesmia (Fabaceae, Muricatae)

Abstract

Background and aims: Adesmia is part of the native flora of Argentina and includes approximately 100 species. Among the species of the genus, A. muricata var. dentata and A. cordobensis var. cordobensis are inermous herbs of the Muricatae series which are widely distributed in northwestern Argentina. Cytogenetic studies are scarce for the genus. In order to carry out a comparative study between these two species, the aim of this work was to provide the first statistically based morphometric studies of karyotypes of A. muricata var. dentata and A. cordobensis var. cordobensis as well as analyze meiotic behavior of them.

M&M: Mitotic metaphases were obtained from roots pretreated with 8-hydroxyquinoline. Chromosomes were stained with 2% propionic hematoxylin. Estimation of pollen grain viability was performed with the Müntzing staining.

Results: All analyzed populations had a chromosome number 2n = 20, symmetrical karyotypes and karyotypic formula consisting of 20 m (metacentric) chromosomes. Adesmia muricata var. dentata and A. cordobensis var. cordobensis have different average chromosome lengths. Furthermore, A. muricata var. dentata presented satellites, not observed in A. cordobensis. In both taxa analyzed, the meiotic behavior was regular, and the viability of the pollen grains was close to 90%.

Conclusions: Chromosome counts, karyotypes, meiotic behavior and viability of pollen grains are novel data. Cytogenetic characteristics allow these related species to be distinguished.
